CHANCE, BWTON, IRVING A. SALKOVITZ, AND ARISTID G. B. ~VACH. Kinetics of mitochondrial javoprotein and pyridine nucleotide in perfused heart. Am. J. Physiol. 213(l): 207-218. 1972.-Fast activation of mitochondrial electron transport can identify the nature and magnitude of their metabolic energy load. Flash photolysis activation of CO-inhibited cytochrome oxidase in the presence of oxygen initiates oxidation of reduced pyridine nucleotide (PN) and flavoprotein (Fp) and the rate of the response is high at high ADP and Ca 2f levels. The fractional extent of the photolysis response of PN and Fp indicates the fraction of the total mitochondrial population containing cytochrome a&O to which oxygen has diffused at the time of the photolysis flash. This fraction is 4X.5 for PN and Fp responses and is considered to be a useful indicator of the effectiveness of oxygen diffusion in the tissue. Such a rapid response ( <O. 1 set) is not observed in vitro in the presence of ADP + Pi, extramitochondrial Ca2+ concentrations are required. Techniques for the rapid perturbation of the physiological and biochemical states of intact tissues are reviewed and the method of flash photolysis of the mitochondrial cytochrome as-CO compound in the presence of oxygen is described, together with the recording of the oxidation-reduction states of mitochondn’al PN and Fp.

Evaluation of Cardiac Mitochondrial Function by a Nuclear Imaging Technique using Technetium-99m-MIBI Uptake Kinetics
Mitochondria play an important role in energy production for the cell. The proper function of a myocardial cell largely depends on the functional capacity of the mitochondria. Therefore it is necessary to establish a novel and reliable method for a non-invasive assessment of mitochondrial function and metabolism in humans. Although originally designed for evaluating myocardial perfusion, 99mTc...
متن کاملDirect Hydrogen Transfer from Reduced Pyridine Nucleotides to Microsomal Cytochrome b, Reductase*
On the basis of several types of experiments (2-7), a mechanism was proposed (7) for the oxidation of reduced pyridine nucleotides by cytochrome bh reductase (1.6.2.2, NADH-cytochrome bs oxidoreductase). According to this scheme, initially an oxidized flavoprotein-reduced pyridine nucleotide complex is formed involving several nucleotide-protein interactions. This is followed by the formation o...
متن کاملInteraction between ferredoxin and ferredoxin nicotinamide adenine dinucleotide phosphate reductase in pyridine nucleotide photoreduction and some partial reactions. I. Inhibition of ferredoxin nicotinamide adenine dinucleotide phosphate reductase by ferredoxin.
Purified ferredoxin has been shown to inhibit reactions mediated by the flavoprotein ferredoxin-NADP reductase. Ferredoxin inhibits the transfer of electrons from NADPH to ferricyanide (diaphorase activity) to NAD (transhydrogenase) and the photoreduction of pyridine nucleotides during the Hill reaction. On the basis of the kinetics of inhibition, it is suggested that the flavoprotein has two b...

متن کاملFlavin and pyridine nucleotide oxidation-reduction changes in perfused rat liver. I. Anoxia and subcellular localization of fluorescent flavoproteins.
Flavin and pyridine nucleotide fluorescence was continuously monitored from the surface of the hemoglobin-free, perfused rat liver with the use of a new double fluorometer. In response to anoxia or after addition of inhibitors of the respiratory chain, the fluorescence intensity excited at 436 rnk decreased, while that excited at 366 rnb increased, representing reduction of flavin and pyridine ...
